影響MALDI-TOF-MS微生物鑒定的因素有很多。本研究主要從微生物培養基、培養狀态、菌體前那說處理、上樣(yàng)量等方面(mià服公n)進(jìn)行MALDI-TOF-MS操作技術和方法的探讨。研究結果顯示：進(jìn)行MALDI-TOF-MS細菌鑒定，培養時(shí)間在4~24 h之間爲佳；盡量選擇無色、不含鹽的術弟液體培養基；0.1 μL菌液是最低鑒定菌量；上樣(yàng)量對(討公duì)結果無影響；對(duì)于菌體山坐前處理，除按照标準化前處理程序操作外，也可將(jiāng)菌體加說通ddH2O制成(chéng)混懸液進(jìn)行點靶煙好鑒定，這(zhè)樣(yàng)更加簡便。本研究有助于減少MALDI-TOF-MS操作盲目性，提高鑒定質量，利于站學操作的規範化和簡便化。
