2017年12月,上海市一養殖場飼養的豬出現咳嗽、呼吸困難、發(fā)熱及迅速轉歸厭身等病症。將(jiāng)豬鼻拭子接種(zhǒn討看g)雞胚進(jìn)行病毒分離,對(duì)血凝試驗陽飛生性樣(yàng)品在SPF雞胚上進(jìn)一步純化、增殖,分離到1株豬流感資飛病毒(A/swine/Shanghai/1厭地205/2017)。采用RT-PCR對(duì)分離毒株進黑件(jìn)行全基因組擴增測序,利用DNAstar軟件,對(duì刀又)測序基因片段進(jìn)行整個閱機員讀框的核苷酸序列同源性比對(du要年ì)分析,并用MEGA6繪制遺傳進(jìn)化樹并分窗光析氨基酸位點。結果顯示:分離毒株爲H1N1亞型,其8個基答吃因片段均屬于類禽型H1N1進(jìn)化分支,沒(méi)有出家雜現不同基因型流感病毒片段之間的重組;分離株HA蛋白的裂解位點序列爲P微如SIQSR↓G,具有典型低緻病性理用流感病毒的分子特征。本毒株的分離鑒定爲分析我國(guó)大陸地區的豬也雨流感流行狀況和分子特征提供了參考。
