雲南省野豬源豬瘟病毒E2及5'NTR基因序列測定分析

豬瘟(CSF)是由豬瘟病毒(CSFV)感染豬引起(村麗qǐ)的一種(zhǒng)急性、熱性、接觸性傳染病,嚴重危害我國(g和內uó)養豬業健康穩定發(fā)展。本研究采用RT-PC慢月R方法,對(duì)從雲南省甯洱縣2頭病死野豬體内分離到的2株黑校CSFV毒株進(jìn)行了E2及5'NTR基因擴增、克隆及序列術裡測定;采用DNAMAN、MEGA6等分子生物學(xué)軟件,對(du器數ì)測得的序列與國(guó)内外參考毒株進(jìn)頻器行了同源性分析及系統發(fā)育分析。結果顯示:2株野豬源CSF民現V株E2、5'NTR基因同源機煙性分别爲87.2%、100%,與國(gu計熱ó)内外參考毒株同源性分别爲81.0%~97.6%、93.6%~98得師.5%,與我國(guó)強毒株友輛Shimen株的同源性分别爲81水媽.0%~81.2%、95.6%;2株野豬源CSFV毒株E2基因屬習風于基因2.1亞型,5'NTR基因屬于基因2.坐用3亞型。氨基酸序列分析顯示:其中一株分離毒株的E2基因有6個氨基酸具有理劇2.1d分子變異特征,另一株兼有2.1d和2.1b亞型分支特征鐵窗,可能(néng)是2.1b和2.1d亞型的過(guò)渡毒株。結年醫果表明,雲南省野豬源CSFV雖存在一定的遺傳衍畫人化,但總體比較穩定。本研究爲雲南省CSF防控提靜著供了分子流行病學(xué)依據,爲進(jìn通動)一步做好(hǎo)分子變異等研究奠定了基礎。
